The Spireites were forced to wait for promotion to the Football League to be secured after falling to a 4-2 defeat to FC Halifax Town at The Shay.
After going ahead through Will Grigg, Chesterfield found themselves 3-1 down by half time.
James Berry pulled one back but that was after Halifax managed to go 4-1 up.
Paul Cook made seven changes to the side that drew to Oldham Athletic on Saturday with Liam Mandeville, Tyrone Williams, Bailey Clements, Ollie Banks, Armando Dobra, Ryan Colclough and Grigg coming into the starting XI.
Andrew Oluwabori tried his luck from 25 yards but found Harry Tyrer difficult to beat as he parried the ball away.
Seven minutes later, Kane Thompson-Sommers did beat the goalkeeper as he slotted the ball goalwards, but Jamie Grimes got back and cleared the effort off the line.
Colclough and Florent Hoti both took turns in shotting just wide of the mark.
Just after the half-hour mark, Colclough cut inside on the edge of the area before striking a post. Grigg was on hand to prod home from a few yards.
Within two minutes, the hosts were level. Rob Harker got in front of Tyrone Williams to turn home Aaron Cosgrave’s cross.
The Shaymen were flying again a few minutes later; Harker was once again put through and he slotted the ball beyond Tyrer.
In added time, Andrew Oluwabori ran through on goal and once again beat Tyrer to put his side 3-1 up at the break.
Berry was brought on just before the hour-mark and almost got one back with a 20-yard drive that drifted wide of the goal.
Clements supplied a teasing cross a couple of minutes later that Grigg latched onto. The striker put it wide from six yards.
Halifax got their fourth and final goal in the 66th minute when Cosgrave was on hand to tap home a cross from five yards.
Grigg was denied by Sam Johnson a few minutes before Berry darted through the home defence and placed the ball past the ‘keeper.
Substitutes Ash Palmer and Joe Quigley also went close but couldn’t quite force the ball over the line.
Chesterfield: Tyrer, Mandeville, Williams (Palmer 76), Grimes ©, Clements, Naylor, Banks, Hobson (Quigley 82), Dobra, Colclough (Berry 59), Grigg.
Unused substitutes: Jones, Jacobs.
